Nucleic acid is a unique functional molecule, which could be designed so as to bind with any given target sequence based on complementarity. Chemically synthesized and modified DNAs are widely used as the genetic probes and regulators. Besides its main role as storage of genetic information, certain DNA exhibits special functions that are very useful in various scenes in molecular engineering. DNA aptamer is a class of the DNA molecules that bind with specific molecules. It is used as the interface between nucleic acids and other biomolecules. DNAzyme is the catalyst that accelerates specific chemical reactions. Elaborate DNA circuits have been designed and used for signal amplification, in which autonomous succession of strand exchange exponentially proceeds after the addition of a trigger.

Targeting a goal of in/on-cell chemistry, we have been constructing several molecular systems by dynamic programming of the functional DNAs and the DNA conjugates. For this goal, 1) we synthesize functional DNA conjugates by covalently combining DNAs and the molecules with desired functions. 2) Preliminary studies on molecular recognition and their incorporated function are carried out using physicochemical and biological methods. 3) Then, finally, their rationally combined systems are subjected to analyses for various biomolecules and regulation of gene expression. The basic techniques used in our research are organic synthesis, physical chemistry, analytical science, and biochemistry. The main fields to output our research are genome chemistry, chemical biology, and bioanalytical chemistry.
